If you submitted your EOI offshore, then you have to update it to onshore once naka SV ka na. Other states have different nomination criteria for onshore candidates. You need to check next FY's listings.
You can't work full-time as of this moment if you are in SV. Per immi's website, up to 48hrs fortnightly only.
